چکیده

to analyze their intrinsic mechanisms. Easy observation of chlorophyll facilitates the study of the underlying mechanisms in chloroplasts, but not in other plastid types. Here, we constructed a transgenic plant enabling visualization of plastids in pollen grains. Combination of a plastid-targeted fl uorescent protein with a pollen-specifi c promoter allowed us to observe the precise number, size and morphology of plastids in pollen grains of the wild type and the ftsZ1 mutant, whose responsible gene plays a central role in chloroplast division. The transgenic material presented in this work is useful for studying the division mechanism of pollen plastids.

Model system for plant cell biology: GFP imaging in living onion epidermal cells.

The ability to visualize organelle localization and dynamics is very useful in studying cellular physiological events. Until recently, this has been accomplished using a variety of staining methods. However, staining can give inaccurate information due to nonspecific staining, diffusion of the stain or through toxic effects. The ability to target green fluorescent protein (GFP) to various organ...

Chimeric green fluorescent protein as a tool for visualizing subcellular organelles in living cells

BACKGROUND It has recently been demonstrated that the green fluorescent protein (GFP) of the jellyfish Aequorea victoria retains its fluorescent properties when recombinantly expressed in both prokaryotic (Escherichia coli) and eukaryotic (Caenorhabditis elegans and Drosophila melanogaster) living cells; it can therefore be used as a powerful marker of gene expression in vivo. The specific targ...

Methods for imaging and analyses of intracellular organelles using fluorescent and luminescent proteins.

One of the important features of eukaryotic cells is the presence of intracellular compartments, organelles. Each organelle plays an essential role in the organelle-specific function; each cooperates with others for maintaining the vital activity of living cells. To analyze the organelle-specific functions, researchers have used many fluorescent and luminescent reporter proteins extensively. Th...

Nonperturbative chemical imaging of organelle transport in living cells with coherent anti-stokes Raman scattering microscopy.

Nonperturbative monitoring of intracellular organelle transport in unstained living cells was achieved with coherent anti-Stokes Raman scattering (CARS) microscopy. To avoid possible interference with the organelle transport introduced by laser radiation, we first examined different illumination conditions. Using a new photodamage criterion based on morphological changes of the cells, we determ...
